The Thermo Scientific ToxInsight Developmental Neurotoxicity Panel (DNTP) offers a complete solution for companies needing an in vitro pre-screen for evaluating large number of compounds for general toxicity and neurotoxicity characteristics through the measurement of multiple biomarkers in individual cells. Traditional methods of assessing developmental neurotoxicity (DNT) require expensive and time-consuming in vivo tests. The predictive, animal-free, automated imaging approach, allows physiologically relevant assays to be utilized earlier in the screening process, saving time, expense and animal usage.

The Thermo Scientific ToxInsight IVT Platform, coupled with the ToxInsight DNTP is the only proven method for analyzing the effect of industrial chemicals on neuronal morphological chemicals in food, consumer products and the environment. The widespread usage of these chemicals has led to fears of a link to the increased incidence of neurological disorders in children. The assay uses NS-1 cells, a subclonal line of rat pheochromocytoma (PC-12) cells, which differentiate forming neurites when stimulated with nerve growth factor (NGF). Automated image cytometry allows quantitative morphological measurements of the fluorescently stained NS-1 cells to determine potential effects on neurodevelopment.

Chromosome mutations of both structure and number are implicated in many human diseases. There is substantial evidence that chromosome mutations and related events in oncogenes and tumor suppressor genes of somatic cells are involved in the induction and/or progression of some types of cancer in human and experimental animals. Thus, the purpose of both the in vitro Thermo Scientific ToxInsight Micronucleus and Comet Genotoxicity Panel Assays are to detect agents that modify chromosome structure and segregation, and through automated imaging analysis, using less compound, time, and overall costs associated with in vivo testing.

The Thermo Scientific ToxInsight Micronucleus Genotoxicity Panel (MNGP) is a standard genotoxicity assay used to assess the potential for compounds to produce chromosomal aberrations. Manual scoring of the micronucleus assay is labor intensive, therefore automation of this task is desired. The ToxInsight MNGP allows for automated and rapid quantification of binucleated cells and micronuclei required for the assay. In the presence of a genotoxic agent (with or without metabolic activation), whole chromosomes or broken DNA may be left behind as micronuclei. The assay automatically analyzes each image set and applies the criteria for targeted cells and micronuclei consistently, ensuring a non-biased analysis. Addition of cytochalasin B improves the assay by allowing you to target only those cells that have successfully reached the binucleate phase, thus allowing distinction between cytotoxic and genotoxic agents. An optional third marker can be used to detect cells with compromised plasma membranes.

The Thermo Scientific ToxInsight Comet Genotoxicity Panel (CGP) is an automated tool for imaging, analyzing and reporting DNA damage by acute exposure to chemicals of radiation. The simple, fast, validated, imaging approach is used to measure Comet formation in vitro in a single channel assay.
